What is checked
Title and description are what people see in Google results. Open Graph and Twitter Card are the title, description and image shown when a link is shared in social networks and messengers. Canonical protects against duplicates, robots and robots.txt decide whether the page gets into search, and schema.org markup helps get a rich snippet with stars, a price or questions.
Why the preview may differ
Google sometimes rewrites the title and description for the query, and social networks cache the image for several days. The preview shows what the page serves right now: if something is wrong, fix the tags and refresh the cache in the social network's debugger.

No, the check reads the HTML the way most search robots see it on the first visit. If tags are added by a script after loading, some systems will not see them - it is better to serve them right in the HTML.
